Protocol-03
Amber PRP Method – 8 Mins

Time

Amber PRP is obtained in 5 steps  and takes around 8 minutes to prepare, the buffy coat forms after this time from which the Platelets will be collected.

Content

Amber PRP is a form of Leukocyte Rich PRP, therefore the final product contains a relative amount of Leukocyte.

Activation

To release growth factors, platelets have to become activated. Activating the platelets inside the body does not require the use of an activator.

Protocol-03
Amber PRP Method

Amber PRP is a Leukocyte Rich PRP preparation approach, this method has been developed to accommodate the needs of users who want Leukocyte in their final PRP milieu.

01

Harvesting the anticoagulant

Anticoagulation is essential for maintaining the fluidity of extravascular blood; after considering which anticoagulant you want to chose, collect it into the syringe.

Process Steps

• Attach a 21 G needle to the NEXT PRP Syringe.
• Harvest 1ml of Sodium Citrate into the NEXT PRP Syringe

02

Harvesting the whole blood

It’s a procedure in which a needle is used to take blood from a vein. Also called phlebotomy and venipuncture.

Process Steps

• Harvest the venous blood from the patient up to 10 ml.
• Close the lid of NEXT PRP syringe.
• Remove the plunger from the piston.
• Turn the syringe upside-down gently in order to mix the whole blood and sodium citrate.

03

Centrifugation

Centrifugation or Fractionation is a separation process in which the blood is divided during a phase transition, into a number of smaller quantities.

Process Steps

• Put the NEXT PRP syringe into the centrifuging machine as the lid will be on top.
• Counterbalance the centrifuging buckets.
• Centrifugate with a swing rotor centrifuge at 830G for 8 mins.

04

Discarding the PPP Layer

PPP stands for Platelet Poor Plasma, it’s blood plasma with very low and insginificant number of platelet therefore we first need to get rid of it before we can collect the PRP.

Process Steps

• Take the syringe from the bucket, remove the lid, affix the connector and plunger.
• Take another luer-lok type syringe and affix it to the connector.
• Discard the PPP layer into the other syringe through the connector until 2ml of plasma is left in the Next PRP Syringe.
• Detach the other syringe from the connector.

05

Harvesting the PRP

Once the blood fractionates into different layers we need to isolate and recover the PRP which contains the growth factors.

Process Steps

• Take another luer-lok type syringe and affix it to the connector.
• Discard the PRP layer into the other syringe through the connector until very little amount of RBCs are presented in the PRP.

06

Final Product

The growth factors are now ready to be injected into the desired areas, your doctor will determine where it is to be introduced.

Process Steps

• Affix desired needle and the PRP is ready to inject.
• 2cc, Leukocyte Rich PRP, 4-5X Fold Enrichment.
